Learning Outcomes
At the end of the course, students will be able to: 1. Understand and discuss the importance and the application of the molecular and biotectechnological approaches in plant breeding. 2. Understand the importance of asexual micropropagation that have breeding aplications 3. The recognazed the differences between conventional plant breeding and aplication of genetic engineering

Course Content (Syllabus)
1st section Laboratory Rules: (laboratory police, machine instructions / any mechanical or electrical apparatus/, preparation of solutions, protocols) 2nd section Isolation of plant and plasmid DNA (material and methods) 3rd section Measure Optical Density of DNA ,DNA cutting with restriction enzymes 4th section DNA Gel electrophoresis: preparation of agarose gel, separation and analysis of macromolecules DNA 5th and 6th section Polymerase Chain Reaction (PCR): Methods, DNA polymerase, oligonucleotide primers. Gel electrophoresis is performed for analytical purposes, often after amplification of DNA via PCR 7th section In vitro culture and regeneration of explants: Methods and conditions, axilary shoot production, tissue culture applications in plant breeding 8th section Genetic engineering in plants: genetic transformation via Α. tumefaciens and Gene gun technique and conditions,regeneration of transformants in selection media , confirmation of transformation 9th and 10th section Chromosome (cytogenetic) analysis: the study of the structure and function of the cell, especially the chromosomes, analysis of G-banded In Vivo Pollen Viability and Pollen Germination 11th section Field trip: Application of Plant breeding methods in self and cross-fertilization plants. Genotype evaluation and selection of desirable traits 12th and 13th section Crossing techniques
